Physical maps in barley
For construction of physical maps in barley, translocation breakpoints in each of the seven chromosomes involving 120 translocations were used as physical landmarks. Since each chromosome was involved in several translocations, it could be divided in sub-regions on the basis of breakpoints, in a manner analogous to that used in the deletion stocks of bread wheat. STS primers from >300 genetically mapped RFLP markers were prepared, and were used with microdissected chromosomes carrying translocated segments as templates for PCR reactions.

This allowed identification of markers flanking each translocation breakpoint, leading to the preparation of physical maps.

Physical map in sorghum

An integrated map of sorghum genome was also published in the year 2000. For this purpose BAC pools were screened with multilocus markers like AFLPs and MITES.
