Genomic-wide physical map in indica rice
In the year 2001, in an indica rice, a total of 21,087 BACs covering about seven times the rice genome were used for the preparation of a genome-wide physical map. The BAC fingerprints were obtained due to restriction digestion and fractionation on an automatic sequencer. The resulting fingerprints were scanned into image files, edited and created into FPC (fingerprint contig) database. Using FPC program, the overlapping clones were assembled into 585 contigs, each contig having 5-128 BACs (the contigs having less than 5 BACs were rejected). Two contigs were treated as overlapping, if the end clones of these contigs had 10 or more common bands. Using this criterion, extended contig's were developed and the number of contigs was reduced from 585 to 298, which covered 97% of the genome. The BACs of each contig were screened using DNA markers, and their correctness validated. In future, this physical map will certainly prove useful for genomics research in rice and other grass species
