Perturbation Theory
A perturbation theory for protein engineering has been formulated, which will enable us to calculate the effects of small changes in amino acid sequence accurately. With the availability of such a theory, there will be no need to perform experiments at each step of protein engineering process to find out the outcome (changes in function) of suggested modifications.
Such a theory will result form the available methods for (i) protein structure refinement and (ii) calculation of theoretical structure of a protein which will be homologous to other proteins of known structure. This perturbation theory can then be tested by using families of monoclonal antibodies which differ only slightly in amino acid sequence. Once tested for its accurate prediction, it can be used for engineering of a variety of proteins.
