Surface Enhanced Laser Desorption-Ionization (SELDI)

SELDI mass spectrometry is an ultra high throughput screening (uHTS) system for protein profiling of extracts obtained from the cells, tissue or physiological fluids. The patented SELDI protein chip system is a unique combination and miniaturization of technologies on a single, unified platform. This system enables capture, purification, analysis and processing of proteins from complex biological mixtures directly on protein chip array surface. Detection of the purified proteins is achieved by laser desorption/ionization time of flight mass spectrometric analysis. In this method, different selective surfaces such as cation or anion exchange surfaces, hydrophobic surfaces, metal binding surfaces, antibodies for specific proteins and DNA can be used on protein chip array to capture proteins and to study protein-protein interactions and protein-DNA interactions to identity proteins.

The SELDI process includes following steps: (i) The sample is applied on the selective surface of the protein chip, so that a subset of proteins is specifically bound to the protein chip. (ii) an energy absorbing molecular assembly (EMA) solution is added on the top of each spot to facilitate desorption and ionization (this solution works like matrix in MALDI). (iii) The chip is analysed by LDI-TOF-MS, which generates a protein spectrum of the different molecular masses present in the protein chip. (iv) The data is transferred to a specialized software package to facilitate data collection and further analysis.

This system can detect and accurately calculate the mases of compounds ranging from small molecules and peptides of less than 1 kDa to proteins of 500 kDa or more.
