Temperature modulated heteroduplex analysis (TMHA) using dHPLC WAVETM system.

TMHA has an extraordinary sensitivity for distinguishing heteroduplexes from homoduplexes, and this principle is utilized for detection of SNPs. The mixture of wild type and variant DNA (SNP mutant) is heated and cooled again, so that the sample will hen have a mixture of homo-and heteroduplexes.

The heteroduplexes partially denature and can then be distinguished from corresponding homoduplexes by ion-pair reversed-phase liquid chromatography (IP-RP-HPLC) used in WAVETM system developed by Transgenomics for distinguishing heteroduplexes from homoduplexes.
