A novel strategy was also developed which combines several advantages of oligonucleotide fingerprinting with RAPD-PCR and microsatellite primed PCR (MP-PCR). In this approach, genomic DNA is amplified with either a single arbitrary 10-mer primer or with a microsatellite-complementary 15-mer or 10-mer primer (as in MP-PCR analysis) and the PCR products are electrophoresed, blotted and hybridized to a γ32P or digoxigenin-labelled SSR [eg. (CA)8, (GA)8, (GAC)5, (GTG)5, (GCGT)4].

Subsequent autoradiography reveals reproducible, probe dependent fingerprints, polymorphic at the intraspeific level. This provides for speed of the assay along. This provides for speed of the assay along with high sensitivity, so that high level of polymorphism is detected. The method was variously termed as RAMPO (random amplified microsatellite polymorphism), RAHM (random amplified hybridization microsatellites), or RAMS (randomly amplified microsatellite sites).
