Phagemids pUC118 and pUC119

This is a pair of phagemid vectors derived from pUC18 and pUC19 respectively and contain cis acting sequences for (i) initiation and termination of phage M13 DNA synthesis and for (ii) packaging of DNA into phage particles. These vectors accept segments of foreign DNA and can be propagated as plasmids in a conventional way.

When cells harbouring these plasmids are infected with a suitable filamentous phage, copies of only one strand of the plasmid DNA are synthesized and packaged into progeny phage particles. Single-stranded DNA can be isolated from the phage particles and used for a variety of purposes including DNA sequencing.
