pGEM-3, pGEM-4, pGEM-3Z, pGEM-4Z

The vectors pGEM3 and pGEM4 carry

(i) ampliciin resistance gene,
(ii) a multiple cloning site (MCS) and
(iii) SP6 and T7 RNA polymerase promoters flanking the cloning site.

In pGEM4, the multiple cloning site is in an orientation, opposite to that in pGEM3. The vectors pGEM-3Z and pGEM-4Z carry lacZ gene HincII fragment encoding α-peptide and the same multiple cloning site that are found in pGEM-3 and pGEM-4, but T7 and SP6 promoters for RNA polymerase are present at the opposite ends of the multiple cloning site in these two vectors.

In pGEM-3Z f(-) additional sequence from the intergenic region of single stranded phage f1 are available, so that this will behave as a phagemid
