Hosts for M13 vectors.

For M 13 vectors, a number of host strains have been recommended. Since M13 enters the host through sex pili, encoded by F factor, only male bacteria carrying F1 episomes are used. These hosts carry a number of genetic markers including the following:

(i) lacZM 15, a deletion in lacZ gene sequence coding for - NH2 terminal region of  galactosidase; this defective lacZ gene is sometimes carried, on F1 episome;

(ii)  (lac-pro-AB), a deletion causing loss of ability to use lactose and needing proline for growth;

(iii) lacIq causing overproduction of lac repressor, which minimizes the production of toxic proteins encoded by foreign DNA under the influence of lac promoter;

(iv) supE, a suppressor mutation that allows the growth of M13 vector carrying an amber mutation (M 13 vectors were earlier but not now required to carry an amber mutation as a biosafety guideline).
