Development of lambda (λ) phage vectors.


There are two classes of lambda cloning vectors:

(i) insertion vectors, which accept inserts, upto 12kb long at a single multiple cloning site (MCS) (e.g. λ gt vectors and AZAP vectors); and

(ii) replacement vectors, which accept inserts, 9-23kb long, involving replacement of a nonessential part (stuffer) of genome (e.g. l FIX vector, λ DASH vector, EMBL series; Charon series); they have a pair of sites flanking a segment of nonessential DNA that can be cut, removed and replaced by foreign DNA.
